The most commonly used technique is using the Cre-Lox recombination system, described 1995. The enzyme Cre recombinase is a site-specific recombinase, which means it is promoting recombination by recognizing a short and specific nucleotide sequence without any added factor. Cre recombinase is known to be expressed in only a few cell types. Hence the researcher would be able to decide where exactly the gene in question is to be knocked out, thus allowing for a greater degree of freedom and scientific accuracy.
